PREVIOUS AND RIEPILOGO
Rapid chromatographic immunological test for the qualitative detection of multiple drugs and metabolites on surfaces and solids at the following concentrations:
Product not intended for medical diagnostic use.
This test only provides an analytical preliminary result.
It will be necessary to use a more specific alternative chemical method to obtain an analytical confirmation result.
The medical judgement and professional opinion will be necessary to evaluate each drug test from abuse, especially when these indicate positive preliminary results.
SUMMARY
Screen Droga Test is a quick test of surfaces or solids that can be performed without instrumentation.
The test uses monoclonal antibodies to selectively detect high levels of specific drugs on surfaces and solids.
Amphetamine (AMP)
Amphetamine is a substance classified in Schedule II of the American DEA sold on prescription (Dexedrin) and also available illegally on the market. Amphetamines are a class of powerful sympatomic agents with therapeutic functions. They are chemically linked to two naturally produced catecholamines from the human body: epinephrine and norepinephrine. High acute doses induce a central nervous system stimulation and cause euphoria, lucidity, decrease appetite and a sense of greater energy and power. Cardiovascular reactions to amphetamines include increased blood pressure and heart arrhythmias. Acute reactions cause anxiety, paranoia, hallucinations and psychotic behavior.
Cocaine (COC)
Cocaine is a powerful stimulant of the central nervous system and a local anesthetic. Initially it causes extreme energy and restlessness which gradually become tremor, hypersensitivity and spasms.
Cocaine, taken at high doses, causes fever, loss of sensitivity, difficulty breathing and loss of consciousness.
Cocaine is often self-administrated for nasal inhalation, intravenous injection and free smoking.
Marijuana (THC)
THC (Δ9-tetrahydrocannabinol) and the main active ingredient of cannabis (marijuana).
If smoked or administered orally, THC produces euphoria.
Who makes use of it presents short-term memory damage and slowness in learning.
You can also experience passenger episodes of confusion and anxiety.
The relatively heavy and long-term use can be associated with behavior disorders.
The peak effect of marijuana administered for smoking occurs within 20-30 minutes and duration and 90-120 minutes after a cigarette.
Metamfetamine (MET)
Metamfetamine and a stimulant drug that energizes some brain systems.
Metamfetamine is very similar chemically to Amphetamine, but its effects on the central nervous system are greater.
Metamfetamine is produced in illegal laboratories and has a high potential for abuse and dependence.
Drugs can be taken orally, injected or inhaled.
High acute doses induce excessive stimulation of the central nervous system, euphoria, lucidity, reduced appetite and a sense of greater energy and power.
Cardiovascular reactions to Metamphetamine include increased blood pressure and heart arrhythmias.
Acute reactions cause anxiety, paranoia, hallucinations, psychotic behavior, and finally depression and fatigue.
Ecstasy (MDMA)
MetilenediossiMetamfetamine (ecstasy) is a synthetic drug first synthesized in 1914 by a German pharmaceutical company for obesity treatment.
Those who take it frequently have experienced side effects, such as increased muscle tension and sweating.
MMDMA is not clearly a stimulant, although it has in common with Amphetamine the ability to increase blood pressure and heart rhythm.
MMDMA produces some changes in perception by increasing sensitivity to light, difficulty concentration and blurred vision in some subjects. It is thought that its mechanism of action takes place through the serotonin neurotransmitter.
MMDMA can also release dopamine, although general opinion is that this is a secondary effect of drugs (Nichols and Oberlender, 1990).
The main effect of the MMDMA, which is likely to have taken a reasonable dose, is to produce a contraction of the jaws.
Morfina (MOP)
The term Oppiaceo refers to every substance derived from opium poppy, including natural products, morphine, codeine and semisynthetic drugs such as heroin.
The term Oppioide is more generic and refers to every drug that acts as an opioid receptor.
Opioid analgesics include a large group of substances that control pain by sedating the central nervous system.
High doses of Morphine can produce high levels of tolerance, physiological dependence and can lead to the abuse of the substance.
Morphine is eliminated without being metabolized and is also the main metabolic product of codeine and heroin.
Ketamina (KET)
Ketamina is a dissociative anesthetic developed in 1963 to replace the PCP (Fenciclidine).
While Ketamine is still used in human and veterinary anesthesia, it is increasingly abused as a drug.
Ketamina is molecularly similar to PCP and thus creates similar effects including numbness, loss of coordination, sense of invulnerability, muscle stiffness, aggressive/violent behaviour, confused speech or afasia, sense of exaggerated force and vacuous look.
It has a depression of the respiratory function but not of the central nervous system and cardiovascular function is maintained.
PRINCIPLE
During the test the sample migrates upwards by capillary action.
A drug, if present in the sample below the limit-limit concentration, will not saturate the binding sites of this specific antibody.
The antibody then reacts with the conjugated drug-protection and will appear a colored line visible in the test area for the specific drug tested.
The presence of drugs above the limit-limit concentration will saturate all antibody binding sites.
So the colored line will not form in the test area.
A positive sample will not generate a colored line in the specific strip test area due to the drug competition, while a negative drug sample will generate a line in the test area for the absence of competition.
For procedural control purposes, a coloured line will always appear in the control area, indicating that a correct sample volume has been poured and that the membrane has been funneled.
REASONS
Each test line contains a monoclonal antibody of anti-drug cavia and its drug-protein conjugates.
The control line contains polyclonal antibodies IgG of goat and IgG of rabbit.
PRECAUTIONS
• Disposable.
• Do not touch the free ends of the strips to avoid contamination.
• Do not immerse the support beyond the maximum level indicated.
• Dip the test into the buffer until one or two red lines appear in the reaction zone (~15 seconds).
• Do not pour samples in the reaction zone.
• Samples can be potentially infected. Establish appropriate elimination and manipulation methods.
• Do not use the Multi Drug device beyond the expiration date.
• Do not use the test if the package is damaged.
• Use the test immediately after opening it.
• Please take into account specificity and cross-reactivity for evaluation.
• Keep and carry the test device at 2-30°C.
CONSERVATION AND STABILITY
Store in the original package sealed at 2-30°C.
The test is stable until the expiry date printed on the package.
The test must remain in the sealed packaging until use.
Don't freeze.
Do not use beyond expiration date.
FORNITIAL MATERIALS
• Device.
• Package leaflet.
• Buffer.
MATERIALS REQUIRED BUT NOT PROVIDED
• Sample collection container.
• Timer.
INSTRUMENTS FOR USE
The test device (in sealed packages), samples and controls should be brought to room temperature (15-30°C) before testing.
Do not open the packages until you are ready to run the test.
FOR AREAS
1. Pass with stripes over the surface to be analyzed.
2. Remove the cappuccino of the bottle provided.
3. Pour all the bottle buffer into the cover.
4. Insert the Multi Test slowly and carefully into the cover with the buffer.
5. Wait for the appearance of lines on the membrane and read the results after 5 minutes. Do not interpret the results after 10 minutes.
FOR SOLIDS
1. Open the bottle and insert the solid into the buffer.
2. Close the bottle with droppers and hood. Shake it briefly. Wait 30 seconds.
3. Remove the cappuccino of the bottle;.
4. Pour all the buffer with dissolved substances into the protective cover.
5. Insert the Multi Test slowly and carefully into the protective cover with the buffer.
6. Wait for the appearance of lines on the membrane and read the results after 5 minutes. Do not interpret the results after 10 minutes.
INTERPRETATION OF RESULTS
Negative*: a coloured line appears in the Control area (C) and a coloured line in the Test area (T). This negative result indicates that concentrations in the sample are below limit threshold levels established for a particular drug tested.
*The colored line/e tone in the test zone (T) may vary. The result must be considered negative every time a weak line appears. Positive: a coloured line appears in the Control area (C) and no line appears in the Test area (T). The positive result indicates that the drug concentration in the sample is greater than the limit threshold established for a specific drug.
Nullo: no line appears in the control area (C). The most probable reasons for the lack of control line are an insufficient volume of sample or incorrect procedural techniques. Read the instructions again and repeat the test. If the result is still null, contact the manufacturer.
CONTROL QUALITY
A procedural control is included in the test.
The line that appears in the control area (C) is considered an internal procedural control.
It confirms that a sufficient sample volume has been used, an adequate membrane imbibation and a correct procedural technique.
Control standards are not provided with this kit.
However, it is recommended to test positive and negative controls such as good laboratory practice to confirm the test procedure and its proper functioning.
LIMITATIONS
1. The Rapid Screen Test Droga Test provides only a qualitative preliminary analytical result. It is necessary to use a secondary analytical method to confirm the result. The recommended confirmation method is mass gaschromatography/spectrometry (GC/MS).
2. A negative result may not necessarily indicate a drug-free sample. You can get negative results when the drug is present below the limit threshold of the test.
3. This test does not distinguish between drugs from abuse and some drugs.
ATTESI VALUES
The negative result indicates that the drug concentration is below the level of detection.
A positive result indicates that the drug concentration is above the level of detection.
